We have a large Nintex form attached to a SharePoint 2019 list that multiple users need to go in and make edits - but when two people are in the form at the same time someone’s changes will not be saved. Is there a way to lock it down so only one person can have the form open at a time to avoid these mishaps?

Welcome to the community.
Unfortunately, there is no way to prevent multiple users from having the form open for the same list items.
Document libraries allow you to check out items so that only one user can edit at a time.
