I created a new document library, and everytime I click on K2 Web Designer, I get HTTP 404 error.
Do I have to give the designer specific access rights in order to open it?

Your issue could be due to a known issue with WebWorkflow when using a SubSite and the Site Collection is using a managed path.
e.g. "http://moss.server.com/ManagedPath/SiteCol/SubSite/DocLib"
A fix for this is currently in development and will be released as soon as possible and currently the only workaround will be not to use a Subsite when your Site Collection is using a managed path.
