Adding Destination Rule to an activity from .NET

Short answer - No.

But if you elaborate on what exactly it is you would like to achieve, maybe we can solve the problem differently.

Remember: Don't confuse a Destination Rule with Destination Users

My requirement is to assign a group as the Destinaion Rule to an activity from Front End. Usually this is done in K2 Studio. But I wanted to know Can I add it from the front end?

I guess it depends how dynamic the requirement is.   If it's a known set of destination groups then in Destination Rules you can define rules on when to route to certain group based on certain criteria (say a field), then in the front-end you can modify this field.
