Within Nintex Designer I am trying to create some additional columns. I click on Create Column and receive the error that says "refused to connect". It doesn't appear to be a permissions issue as I am owner/administrator of the site and I created the list. Any suggestions would be appreciated.

+17@PLB I've seen this happen from time to time and it was some weird browser thing. If you keep running into that, just close out the designer and create the column the normal way from the list settings option.
This appears to be a bug with Chrome. It has been escalated to Nintex Dev for review. The workaround is to use IE.
