I want to create a SharePoint List Item using a Nintex form and once entered to automatically be uploaded into Excel using a workflow. Is this possible with Nintex? And if so, do you need to have the Enterprise version? Thank you.

Can you explain a bit more about the "automatically uploaded into Excel"?
Do you want to:
a) Create a new excel document using values of the list item?
b) Generate a excel document with all the list values?
c) Something else?
For option A Vadim Tabakman has written a nice article about it
For option B, you could use the Export to Excel function
If you have SHarePoint Entreprise, you have Excel services webservice, so this is your solution :
