we have configured K2SQL authentication and enabled all the Pre-requisite and however when we try to login using K2SQL then getting below error.

Claim mapping configuration cannot be found for this claim. Claim information: Name='XXX', Issuer='FormsSTS', Original Issuer='FormsSTS'. Please ensure that you have configured the K2 server as specified in K2 Help: Installation and Configuration > Configuration > SharePoint > Claims-based Authentication.

I assume the steps below were followed?

https://help.nintex.com/en-US/k2five/icg/5.6/Content/Configure/SF/MultiAuthSQLUM.htm

I would check to make sure the thumbprint for the FormSTS and for the FormsSTS web.config matches:

And that all of the Claims Type Mappings are there:

Double check for any leading/trailing white spaces and miss-spelling, as that can cause Claim mapping configuration to not be found.  Case-sensitivity may need to be considered also.
