I am wanting to create a task for the assignee to simply acknowledge work has been completed, only having one response. I was able to do this in the SharePoint 2013 workflow. Can this be done in O365 workflow?

Have you tried creating a content type that just has the one choice and then use the "Use existing predefined content type" link in the task action and selecting your content type?
