We have used lot of sql request actions in on-premise version of nintex, we are planning to migrate to O365 and i am surprised that there is no sql request action in it. May i know is there any alternative solution or when the sql request action is available in O365?

The equivalent action in Workflow for O365 is called "Execute SQL".
You should configure a VPN between SharePoint for O365 and where the SQL Server is hosted because the credentials are passed as plain text.
