I have Imported the existing workflow to the calendar list on SharePoint O-365 and I want to edit the task form of "Assign a task" action but the button is disabled. Please provide the solution for this.

Nintex Workflow does NOT offer any possibilities to modify forms.

Don't get me wrong but are you sure you have understood that Nintex Forms and Nintex Workflow are two seperate apps to install?
+17You have to add a separate app to a site where you want to edit your forms. The app is called Nintex Forms for Office 365. Go to "Site Contents" and click "add an app", type "forms" and click to add:
After the app is provisioned to the site, the button becomes active 
If you have a license for Nintex Workflow for O365 then Forms are as well included.

We will need more details than we currently have, and a screenshot is a good place to start.
Although you have now indicated to that you have added the O365 Nintex Forms App to your site, that would be the number one reason why this is happening... so we need more in order to help you.
