We are planning to move to SharePoint 2016 from SP2010 this year and as part of that move we will need to move our workflows. What will be the path/process to upgrade from Nintex 2010 to Nintex 2016 as part of this move? Will there be any requirement to have Nintex 2010 at the latest current version before upgrading?

It depends how you will plan to migrate.
It could be SHP2010>>SHP2013>>SHP2016 as recommended by Microsoft. In this case you will need to migrate Nintex the same way 2010>2013>2016
There are some tools like Metalogix that makes a direct migration 2010>>2016. They also have a tool for migrating Nintex 2010>2016
