I see that a K2 workflow can create a document set. Can a K2 SmartForm be used to edit the propties of a document set so that the property fields can pull from another LOB system/SmartObject?

This sounds very possible. You can perform actions on SmartForms via CRUD events to SharePoint list and libraries. I could theoretically load in properties from one or more SharePoint Libraries and update them via control or rule events.
Was there something in particular that you were trying to accomplish?
I enabled Document sets on a test SharePoint library. After refreshing the service I now have the ability to get/update/delete/create:
