License for SharePoint 2016

We have one App server and one front end server. Nintex is counting App server as front end as well. All the sites are hosted on the App server. Can we use single license for our environment ?

A server can be called whatever you like, i.e., web front end, app server, etc.
What determines the function of a server is the services that run on it.
If your “App Server” is hosting all the sites, it runs the Microsoft SharePoint Foundation Web Application service, which, according to Microsoft, is a web front-end service.  All web front ends need to be licensed for Nintex.
