I have added a role to destination user of an activity, and using a cutomize mail in the same activity , in To mail option destination user radio button is disabled. Is there any aprticular setting to send mails to the destination users.

you need to deal with advanced destination (When your are on your Destination Rule screen, click on the previous button and select "Advanced option) in your destination rule... and you just need to choose one of the 2 "Plan per destination" option. Then the checkbox will be enable.
