We are using the K2 worklist web part and one of customization that is available when you select 'Modify this web part' option is 'Maximum Items' and the default value for this 100.
We have a need where we need all the items in the work list.
Will setting this field to 0 or -1 help?

I dont think it is possible. Maximum Items field exist for performance reasons. Imagine all the users trying to retreive all of their workitems first thing in the morning - it might kill the K2 or SQL server. I dont see actual business benefit in listing more than 100 items at a time on a single page.
The webpart sourcecode is available. So you can do the changes as per your requirements.
