After releasing a List/Item the responsible person decided that the single line text column needs to be changed to a multiple line text column. I know this is possible but there are Items already in the List – how will the text be affected that is already in the single line text column? Will the text be removed/deleted? I'm hoping nothing will happen to the text already in the column.

Changing from single line of text to multi line of text (plain text/rich text/enhanced rich text) nothing will happen to data in list.
But you need to change the control (from single line text to multi line text) in Nintex Form and Need to cross verify steps in workflow.
But i suggest always it is better to create a new multi line text column and copy data from single line text to multi line text using a simple workflow.
