I would suggest opening a support ticket with Nintex support to help you walk through troubleshooting the website and IIS application settings. If you are the web site admin for your organization, you can work with support to look at the configuration and error log messages. If you're not the admin, you may want to have your website admin review the IIS settings and error log for that site.
